P9WII3 | Reviewed | Endoribonuclease MazF6 | Mycobacterium tuberculosis (strain ATCC 25618 / H37Rv). | 35357163 | Functional and Biochemical Characterization of the MazEF6 Toxin-Antitoxin System of Mycobacterium tuberculosis. | [Function][Expression] | Protein/gene_name: MazF6; mazF6. Function: Analysis of paired-end RNA sequencing data revealed that the over expression of MazF6 resulted in differential expression of 323 transcripts in M. tuberculosis. The transcription profiles obtained in M. tuberculosis upon over expression of MazF6, were similar to those obtained from bacteria exposed to stress conditions such as nutrient starvation, enduring hypoxic response (EHR), non-replicating persisters (NRP), and drug-induced persisters. Toxin over expression led to the up regulation of transcripts involved in cell wall and cell processes, intermediary metabolism and respiration. Additional Annotation: From network analysis, it was observed that amongst the top response network hubs, whiB3, a redox-sensitive transcriptional regulator that regulates the expression of several genes encoding polyketide synthases, was identified in the MazF6 overexpression strain. A paired-end RNA sequencing-based method for mapping and quantifying the cleavage of endoribonucleases was applied to map cleavage sites that result from the overexpression of MazF6 by calculation of the mRNA protection ratio in its native host, M. tuberculosis. | Gopinath Chattopadhyay | 0000-0002-6408-1968 | 2022-8-16 |
